The Applied Stem Cell Sciences team at AstraZeneca in Gothenburg, Sweden are looking for our next colleague to join us Senior Research Scientist focusing on in vitro Target Validation to support . In this position you will play an important part in our growing Translational Genomics department. You will be working alongside our stem cell scientists and disease areas specialists. Using your strong knowledge and experience in molecular biology, cell biology, and assay technologies you will have the opportunity to work with cutting edge cellular models of disease to validate various target hypothesis. What you will do: As a Senior Research Scientist you will contribute in this lab-based role and focus on cellular model generation and validation for a range of target biology research programmes. Some of the main responsibilities include culture of primary cells, differentiation of induced pluripotent stem cells (iPSCs), cell line engineering and using these model systems to validate novel targets and mechanisms using a variety of assay technologies. In the daily work you will develop and support a number of disease-relevant cell culture models across a variety of therapeutic focus areas. In collaboration with your colleagues, you are responsible for designing and executing cell-based experiments to validate novel targets using tool compounds and genetic knockdown/gene editing approaches. Along with this you will culture and differentiate hPSC lines to physiologically-relevant cell types in 2D or 3D culture conditions and perform molecular biology protocols in support of in-house quantitative PCR, digital PCR and NGS assays. Essential in the role: * PhD in Developmental Biology, Cell Biology, Gene Editing or a BSc/MSc degree with significant experience. * Experience with the culture of multiple mammalian cell types including primary cells and stable cell line generation. * Demonstrable experience with a variety of molecular biology techniques including CRISPR/CAS9 applications and relevant bioinformatics tools. * Hands on experience with quantitative assay development such as qRT-PCR, ELISA, Western blots and/or cell- based functional/ potency assays. To be successful in the role, you should also have great communication skills with ability to build open and collaborative relationships and enjoy team work as a member of a multidisciplinary team. Forskningen i Läkemedelskemigruppen vid Göteborgs Universitet innefattar datorstödd design, organisk syntes och karakterisering av substanser ämnade att användas som eNeurofarmaceutiska preparat för nervsystemet. I det här projektet är målet att ersätta biokemisk med elektronisk signalering. Traditionella läkemedel baseras på biokemiska principer utan att ta hänsyn till elektroniska egenskaper. Vi fokuserar här på organiska elektroniskt ledande material som självorganiserar sig i eller i närheten av nervsystemet. Målet är att bilda elektroniska kretsar som efterliknar strukturen och organisationen av nervsystemet för att detta skall kunna integreras. Arbetsuppgifter Arbetsuppgifterna omfattar främst design och syntes av småmolekyler för utforskandet av biokompatibla semikonduktorer för integrering med centrala nervsystemet. Fokus för arbetet är organisk syntes och karakterisering men inkluderar också evaluering i in vitro och in vivo system för att fastställa biokompatibilitet och aktivitet av föreningarna. Kvalifikationer Behörig att anställas som forskare är den som har svensk doktorsexamen eller har en utländsk examen som bedöms motsvara doktorsexamen. Vi söker en disputerad organiskkemist med goda kunskaper om och erfarenhet av design och syntes av biologiskt aktiva substanser och ledande polymerer/supramolekylära system, speciellt syntes av monomerer till sådana system. Den sökande ska ha en bred kunskap i organisk syntes och vara väl förtrogen med heterocykelkemi och kol-kol korskopplingar. Då arbetsuppgifterna även inkluderar elektrokemisk karaktärisering av de syntetiserade föreningarna är erfarenhet från cyklisk voltammetri ett krav. What you will do: In this role you'll work as a specialist in the field of Molecular Cell Biology and be responsible for cellular model generation and validation for a range of target biology research programmes. In the daily work, you will independently devise, initiate or contribute to the implementation of project plans to investigate target biology and generate work-plans to agreed timelines to meet project objectives, including delivery of more than one agreed area of work in parallel. You will design and execute cell-based experiments to validate novel targets using tool compounds and genetic knockdown/gene editing approaches. As part of the role you will coach and support junior scientists by advising them and providing expert input in interpretation and presentation of complex data - and provide scientific leadership and represent the group at internal as well as external meetings. We encourage you to drive internalization of emerging technologies and actively contribute to publications in peer-reviewed journals. You are driven to come up with novel approaches and excited about publishing your research in high impact journals. Essential for the role: To be successful in the role you need to have an open and innovative attitude and the ability to interact and communicate efficiently across interfaces of global and scientific functions and fields. Experience of leading collaborative projects and mentoring more junior scientists is also essential. Job assignments Brown and white adipocytes play important roles in regulating metabolism. We have developed both in vitro and in vivo systems to study how metabolic regulation in adipose tissue takes part in regulation of systemic energy turn-over during normal and pathological conditions e.g. insulin resistance. In this project we will focus on how the adipocyte regulate its energy turn-over and substrate preferences during metabolic challenges such as food restriction, cold, and metabolic diseases e.g. type 2 diabetes. Our goal is to gain a detailed understanding of the molecular processes that regulate metabolism in the adipocyte and how this affects other organs and thus take part in regulation of metabolic interplay between different organs to achieve a healthy and sustainable metabolism for the entire organism. Qualifications A PhD degree in a relevant field. The ideal candidate will have experience in molecular regulation of metabolism and the techniques used in molecular biology, biochemistry and animal work measuring metabolic parameters such as glucose tolerance tests and also experience of different methods such as protein expression, cloning, mutagenesis, bioinformatics, and experience of mice work. Experience in the methods used to monitor basic metabolic parameters such as basal and insulin-stimulated glucose uptake and beta-oxidation in adipocytes and muscle cells is important together with specific knowledge of how to combine this with overexpression and knock down, using siRNA and shRNA, of gene expression using inducible virus-mediated expression is especially important for this project. Experience in quantitating translocation of glucose transporter is also an important merit.  Employment The employment is limited (temporary) for 2 years and full-time with placement at the Institute of Biomedicine.
