On behalf of our client Takura, a Swedish biopharma company, we are now looking for a dependable and experienced researcher, preferably within stem cell biology, to manage the company’s preclinical projects. Takura with its subsidiaries is working with MSC treatments (human and veterinary applications) and TIL cancer therapy. Currently, the location of a GMP facility in Stockholm is under consideration. Takura also have a joint venture in Abu Dhabi, UAE. 

Responsibilities

As a preclinical project manager, you will manage Takura's preclinical projects as well as adding to the team as a valuable member in various company projects. You will independently be managing the full scope of the preclinical projects. This will include planning and performing experiments, evaluating and summarizing data, identifying trends and patterns and designing the preclinical studies that will be outsourced. In your role, you will identify and communicate with external preclinical partners to find the best solutions for each project. You will compile and present your reports, findings and/or problems to management and other stakeholders. In this role, it is necessary to continuously follow a strict code of ethics and protect confidential information at all times.
 
You will: 
  • Conducting necessary research on study projects by using books, journal articles, newspaper sources, questionnaires, surveys, polls etc. to gather data.
  • Interpreting study project specifications from external partners and developing a work plan that meets the set requirements.
  • Writing study reports, a preclinical summary of data for clinical documentation etc.
  • Anticipating research issues and promptly resolving them.
  • Writing proposals and delivering oral and written presentations when required.
  • Assisting management with budget and time schedules.
  • Ensuring that required ethical approvals, biobank approval, material transfer agreements etc. are in place for the relevant research.
  • Communicating with sales and technical experts regarding potential purchases of equipment, raw materials etc.

Qualifications

Essential for the role:
  • Minimum a Bachelor's degree in cell biology, immunology, biochemistry or similar relevant area.
  • PhD in stem cell biology, transplantation, immunology or similar relevant topic.
  • >3 years work experience in the relevant area after PhD defense.
  • Work experience as a project leader in the industry or with industrial partners
  • Sound understanding of research methodologies.
  • Proficiency in MS Office.
  • Experienced in working with a laboratory management system.
  • Fluent in English both orally and in writing and preferably also Swedish.

You need to have strong problem-solving skills and a flexible state of mind as projects can change quickly, and you will manage many projects in parallel. Modulation of ER stress responses in primary lung cells

Thesis project:

Endoplasmic reticulum (ER) stress, arising from perturbed protein homeostasis, activates the unfolded protein response (UPR) that leads to a myriad of downstream cellular events. We believe that dysregulation or defective UPR responses plays an important role in chronic respiratory diseases such as COPD or IPF. This project aims to investigate and modulate the UPR pathway activation in primary lung cells of different origin to identify the major cell types targeted by ER stress and its specific downstream UPR responses.

Responsibilities

In this placement, the student will be part of a cross-functional team within Research and Early Development, Respiratory and Immunology (R&I), building scientific knowledge around disease-causing molecular mechanisms and identification of novel targets for treatment. Specifically, the student will be working closely with a team investigating the role of ER stress in COPD and IPF.
 
Objectives and goals:
  1. Establish protocols to induce ER stress and monitor the UPR and associated respiratory disease-relevant biological pathways in primary lung cells.
  2.  Acquire extensive training in fundamental molecular and cell biology techniques, including qPCR, cell imaging, Western blot and other cellular/protein assays. 
  3. Contribute scientifically to the cross-functional ER stress team.
  4. Get insight in project work and drug discovery. 

Qualifications

Essential Requirements

Role Overview

You will play an active role in implementing and executing scientific research and development within the CVRM Metabolism in vitro team, enabling the prosecution of a rapidly growing portfolio of small molecule and biologics drug projects. This is a "hands-on," laboratory based scientific and technical role in our in vitro metabolism team supporting projects from target validation to lead optimisation phases.
Role specifics

This position will be laboratory based and you will be responsible for several technical and experimental processes to support the CVRM Research group.

Responsibilities

  • Responsibility for design, execution and analysis of in vitro and ex vivo assays.
  • Predominantly functional and cellular screens supporting Target Validation, Lead Isolation and Lead Optimisation for the CVRM Research Area.
  • Communicate results, conclusions and recommendations to colleagues, line manager and multi-disciplinary teams.
  • Perform extended and specialized characterization when required for projects.
  • Record experiments in lab books and databases.
  • Play a leading role in assay development and optimization of disease relevant primary and transfected cell assays.
  • You would report to an expert member of staff and would be encouraged to be able to plan your daily work within direction set by your supervisor.
You will:
  • Be asked to provide interpretation of assay results
  • Work within a line and/or matrix/project team.
  • Demonstrates scientific rigor in assessing own data and that of others.

Qualifications

Essential
  • Be a "hands-on" expert with in vitro models and methodologies.
  • Enthusiasm for laboratory work, scientific curiosity
  • Experience in running in vitro biochemical and cellular assays to provide potency and activity readouts, including use of primary and recombinant cell lines.
  • Significant cell culture experience.
  • Experience of working with medium to high density microtitre plate technologies
  • Experience in data analysis and interpretation
  • Experience of working in accordance with established SOPs and codes of conduct.
  • Capability to work seamlessly with cross-functional project teams to provide high quality data in a timely manner.
  • Demonstrable ability to work within a line and/or matrix/ cross-functional project team.
  • Strong interest in providing materials and data for scientifically challenging work
  • Strong organizational, time management and communication skills
  • Strong, demonstrable analytical ability
  • Good oral and written communications skills
Desirable
  • Background in metabolic disease research areas.

Developing a method for characterizing and improving  stem cell differentiation

Introduction
Biopharmaceuticals R&D is now building a new cell therapy department with the goal to establish AstraZeneca as a world-leader in the stem cell field and to build a competitive pipeline within cell therapy across disease areas within Cardiovascular, Renal and Metabolism (CVRM) and Respiratory and Inflammation (R&I). By directing differentiation of pluripotent stem cells into precursor cells or to mature cell types, novel therapies can be developed targeting a wide range of diseases such as heart failure, kidney and liver diseases.

The project aims to develop a novel analytical method that will be used for optimizing differentiation protocols.

Responsibilities

The primary focus of the project will be to culture pluripotent stem cells and differentiate them towards cardiac cells. Samples from the differentiation process will be collected and analyzed, e.g. using ELISA and flow cytometry. 

Qualifications

  • A strong interest in stem cell science.
  • Highly motivated, having a scientific mindset and can easily comprehend and apply ideas from the literature. 
  • Ability to watch & learn, and then perform experiments on your own, with some guidance.
  • Good organizational skills.
  • Written and oral communication skills in English.
  • Cell culture experience.

Clearing for Lung Tissue Transparency – towards 3D imaging

Did you know organs can be made transparent and be imaged in 3D? If you are interested in working with 3D imaging of lung in a group with advanced imaging labeling techniques – please come join the Swedish Imaging Hub at Imaging and data analytics, CPSS (clinical and pharmacological safety sciences) for you master thesis.

Now that we have developed clearing protocols for creating transparent organs a number of questions about exactly what can be viewed, how structures are connected to eachother, in what magnification can the tissues be viewed and how many things can we label and investigate at the same time. The organ we will start exploring this in is the lung. Lung tissues have a complex matrix of heterogenic molecules, for example elastin, collagen and phospholipids in cell membranes. Our clearing method remove many of these heterogenous molecule while proteins and carbohydrates that are covalently bound to the matrix and each other are still preserved, making us able to label what is of interest with fluorescent tags and image them. At first we will focus in imaging a lot of the important proteins naturally occurring in the lung together with inhaled drugs and on top of that explore capturing a detailed 3D view of disease related phenotypes like inflammation and tumors. You will join an organization at AZ focusing on Imaging, imaging analyses and how to use artificial intelligence to support the image analyses. The organization is a part of AZ that works with safety concerns which might arise during drug development.

Responsibilities

By joining our team you will learn techniques and principles around tissue labelling and imaging. You will learn to appreciate how important imaging is during scientific trouble shouting and drug development. The goal of your particular project will be to:

  • Further improve the clearing protocol in particular to speed up the clearing and labeling process.
  • Test different labeling techniques to get the most out of the 3D information as possible
  • Evaluate the 3D high resolution images and support the development of digital image analyses to quantitatively determine numbers and volume measurements
