In this Post-Doc position in Mikael Molin's research group your work will contribute to the understanding of molecular mechanisms of aging and age-related disease. More specifically you will investigate the contribution of peroxiredoxins, conserved antioxidants, signaling modulators and chaperones, to the ability of reduced caloric intake to slow down age-related deterioration in a simple model organism. You will help advance world-leading, state-of-the-art research at the nexus of Molecular Cell Biology and Systems Biology with a large potential for high-impact publications and for follow-up studies in mammals. Information about the division and research The Post-Doc position is placed in the vibrant and successful Division of Systems and Synthetic Biology, that is part of the Department of Biology and Biological Engineering at Chalmers. The research group of Mikael Molin has in later years pioneered the molecular understanding of aging by pin-pointing several key roles of the ROS hydrogen peroxide in signaling impinging on aging. Instrumental effectors of the surprising, beneficial effects of this reactive molecule in slowing down aging are the enigmatic peroxiredoxins. Peroxiredoxins function as antioxidants, signaling modulators and molecular chaperones. The division has an extensive instrument platform for analytics, molecular biology, protein production and purification which are all available for the project. In addition, core facilities with advanced equipment for metabolomics, proteomics and microscopy are available at the department, the campus and the neighbouring University of Gothenburg. Major responsibilities As a Post-Doc you will conduct experimental work and data analysis aiming to clarify the impact of peroxiredoxins on protein synthesis and aging. Major research tasks include surveying the many potential downstream effectors of hydrogen peroxide signaling on protein synthesis revealed by a recent extensive targeting of cysteine-thiols in the protein synthesis machinery (Topf U et al, Nat Commun, 2018), for effects on peroxiredoxin-related traits relevant in aging. This knowledge needs to be integrated with our unpublished global efforts and with ongoing studies investigating the impact of peroxiredoxins on human neurodegenerative disease-relevant proteins. The position includes supervision of undergraduate students to a certain extent as well as local, national and international collaboration. You will be expected to regularly present data in different forums including scientific conferences, meetings and seminars as well as to write manuscripts and apply for (minor) funding. The position is meritorious for future research duties within academia as well as industry/the public sector. Qualifications You should have a PhD in Molecular Cell Biology, Microbiology or a related discipline. Extensive hands-on research experience in the fields of Molecular biology, Molecular Genetics, Cell Biology and Microbiology, are considered an advantage. Special emphasis will be given to qualifications in RNA analysis, studies of protein synthesis, aging and the defence against ROS in yeast, especially regarding the function of redox metabolism and redox signaling in these processes. You need to be motivated, self-propelled, able to withstand stress, quality conscious and possess excellent technical abilities. You should be able to set up new experimental procedures and be good in finding solutions to technical problems. Cooperation is important for the position and therefore good social, interpersonal skills are important. Furthermore, you should have good analytical skills, a keen interest in the interpretation of complex data and an excellent ability to summarize and report results. An ability to work in teams and a drive for own development are also important. Good communication skills in both spoken and written English are a requirement. Ansvarsområden

Tjänsten som laboratorieingenjör är i den operativa sektionen på QC mikrobiologi och innebär att utföra mikrobiologiska analyser på inkommande råvaror, APIer, processprover och färdig produkt samt identifiering av mikroorganismer. I tjänsten ingår också att driva utredningar och förbättringsarbete samt delta i valideringsarbete.

Som laboratorieingenjör är dina huvudsakliga arbetsuppgifter och ansvarsområden:
• Analys av steriltest i renrum (B-klass).
• Inslussning av produkter och material till renrum (från O-klassat till B-klass).
• Analys av koktest, endotoxintest och totalantal mikroorganismer (bioburden).
• Att självständigt utföra det dagliga arbetet som avläsningar av prover, rapportering av resultat, granskning, trendning och beställning av förbrukningsmaterial etc.
• Att självständigt driva avvikelseutredningar inom arbetsområdet (initiera, utreda, dokumentera).
• Att självständigt uppdatera instruktioner och driva förbättringsarbete.
• Utföra identifiering av mikroorganismer samt utföra ankomstkontroller av stammar och substrat.
• Delta i avdelningens valideringsarbete.

Som person bör du vara flexibel, noggrann, ha god kommunikationsförmåga, är nyfiken, målmedveten och strukturerad. Det är viktigt att du har mycket god samarbetsförmåga och tar ansvar så att gruppen levererar enligt satta tidslinjer och kvalitetskrav. Du behöver ha förmåga att kunna driva administrativa uppgifter parallellt med analysarbete och anpassa ditt arbete efter de prioriteringar som gäller. Du är också självgående i ditt arbete, tar egna initiativ och påvisar gott omdöme. I rollen agerar du utifrån de riktlinjer och krav som styr arbetet inom läkemedelsindustrin, det krävs att du strukturerat och noggrant utför och dokumenterar dina arbetsuppgifter.

Arbetstider

Kvalifikationer

• Akademisk utbildning inom bioteknik, biologi, mikrobiologi eller annan motsvarande utbildning.
• Erfarenhet av mikrobiologisk kvalitetskontroll och analysarbete.
• God förmåga att utrycka dig på engelska och svenska i tal och skrift.

• Erfarenhet av sådant arbete inom läkemedelsindustrin är meriterande.
• Erfarenhet att ha arbetat med renrumsarbete i B-klass, analyserfarenhet av steriltest, endotoxintest, bioburdentest, identifiering och/eller mikrobiologiska utredningar är meriterande.

Modulation of ER stress responses in primary lung cells

Thesis project:

Endoplasmic reticulum (ER) stress, arising from perturbed protein homeostasis, activates the unfolded protein response (UPR) that leads to a myriad of downstream cellular events. We believe that dysregulation or defective UPR responses plays an important role in chronic respiratory diseases such as COPD or IPF. This project aims to investigate and modulate the UPR pathway activation in primary lung cells of different origin to identify the major cell types targeted by ER stress and its specific downstream UPR responses.

Responsibilities

In this placement, the student will be part of a cross-functional team within Research and Early Development, Respiratory and Immunology (R&I), building scientific knowledge around disease-causing molecular mechanisms and identification of novel targets for treatment. Specifically, the student will be working closely with a team investigating the role of ER stress in COPD and IPF.
 
Objectives and goals:
  1. Establish protocols to induce ER stress and monitor the UPR and associated respiratory disease-relevant biological pathways in primary lung cells.
  2.  Acquire extensive training in fundamental molecular and cell biology techniques, including qPCR, cell imaging, Western blot and other cellular/protein assays. 
  3. Contribute scientifically to the cross-functional ER stress team.
  4. Get insight in project work and drug discovery. 

Qualifications

Essential Requirements
  • Good networking skills and interpersonal skills.
  • Interest and curiosity in molecular biology and the experimental work behind drug discovery.

  • Understanding of cell and molecular biology.

Desirable Requirements

Cardiovascular Renal and Metabolism (CVRM) Research – Metabolism In Vitro Team - Gothenburg

Role Overview

You will play an active role in implementing and executing scientific research and development within the CVRM Metabolism in vitro team, enabling the prosecution of a rapidly growing portfolio of small molecule and biologics drug projects. This is a "hands-on," laboratory based scientific and technical role in our in vitro metabolism team supporting projects from target validation to lead optimisation phases.
Role specifics

This position will be laboratory based and you will be responsible for several technical and experimental processes to support the CVRM Research group.

Responsibilities

  • Responsibility for design, execution and analysis of in vitro and ex vivo assays.
  • Predominantly functional and cellular screens supporting Target Validation, Lead Isolation and Lead Optimisation for the CVRM Research Area.
  • Communicate results, conclusions and recommendations to colleagues, line manager and multi-disciplinary teams.
  • Perform extended and specialized characterization when required for projects.
  • Record experiments in lab books and databases.
  • Play a leading role in assay development and optimization of disease relevant primary and transfected cell assays.
  • You would report to an expert member of staff and would be encouraged to be able to plan your daily work within direction set by your supervisor.
You will:
  • Be asked to provide interpretation of assay results
  • Work within a line and/or matrix/project team.
  • Demonstrates scientific rigor in assessing own data and that of others.

Qualifications

Essential
  • Be a "hands-on" expert with in vitro models and methodologies.
  • Enthusiasm for laboratory work, scientific curiosity
  • Experience in running in vitro biochemical and cellular assays to provide potency and activity readouts, including use of primary and recombinant cell lines.
  • Significant cell culture experience.
  • Experience of working with medium to high density microtitre plate technologies
  • Experience in data analysis and interpretation
  • Experience of working in accordance with established SOPs and codes of conduct.
  • Capability to work seamlessly with cross-functional project teams to provide high quality data in a timely manner.
  • Demonstrable ability to work within a line and/or matrix/ cross-functional project team.
  • Strong interest in providing materials and data for scientifically challenging work
  • Strong organizational, time management and communication skills
  • Strong, demonstrable analytical ability
  • Good oral and written communications skills
Desirable
  • Background in metabolic disease research areas.
